We isolated an α-amylase inhibitor( AI) from transgenic field pea in which a Tendergreen α-amylase inhibitor gene was transduced and the sugar chain structures of AIs from transgenic field pea and Tendergreen bean were compared. Plural electrophoretic bands were found in each AI and each subunit of each AI and these electrophoretic patterns were different between transgenic field pea and Tendergreen. However, each subunit with the deglycosylation using TFMSA showed single band. The structure of a major sugar chain of α subunit of Tendergreen AI was M9. Structures of major sugar chains of α subunit of transgenic field pea AI were M8 and M9. On the other hand, major sugar chains in the βsubunits were the same, but the compositions were slightly different. Transgenic field pea AI and Tendergreen AI have heterogeneity in the composition of the binding sugar chains. |
