The stability of the α- amylase inhibitor（TAI）from Toramame（Phaseolus vulgaris L.）toward the attack of proteolytic enzymes（pepsin, trypsin and chymotrypsin）was examined by estimating the inhibitory activity, SDS-PAGE and immunoblot of the digests. TAI showed strong resistance against pepsin or trypsin, and was not damaged. On the other hand, TAI was unstable toward chymotrypsin attack to cause the gradual loss of the α- amylase inhibitory activity. The presence of BSA in the digestion mixture prevented the TAI degradation and was accompanied with a protection of the activity. These findings suggest that TAI is protected to some extent against chymotrypsin digestion when ingested with a food protein. |
