Four proteins (BCP -IA, -IB1, -IB2 and -II) having chitin-binding ability were purified from seeds of buckwheat (Fagopyrum esculentum cv Shinano No.2) by affinity chromatography, ion-ex-change chromatography, and reverse-phase HPLC. All these proteins had similar amino acid compositions characterizad by high contents of half-cystine and glycine corresponding to about 50% of total component amino acids. Binding experiments of BCP-IB1 to chitin at 30℃, pH 8.0 indicated that the equilibrium absorption constant, Ka, and maximum amount of the bound protein, [PC]max, were 0.55×10^4 M^<-1> and 17.1 μmol/g, respectively. BCP-IB1 also showed the growth inhibition against Saccharomyces, Candida, Penicilium, and Aspergillus tested. |
