An α-amylase inhibitor (DAI) from Daifuku bean is composed of two kinds of glycopolypeptide subunits, α and β. Sugar compositions of DAI and its subunits α and β were shown to consist of mannose, N-acetylglucosamine and xylose. These results indicated that DAI had asparagine-linked sugar chains. Asparagine-linked sugar chains of DAI and its subunits α and β were liberated by hydrazinolysis. After N-acetylation and pyridylamination, structures of pyridylaminated sugar chains were estimated by size-fractionation HPLC. Several kinds of pyridylaminated oligosaccharides were obtained from DAI and its subunits α and β. Structures of major sugar chains of subunit α were M6 and M9 of high mannose-type oligosaccharides. The structure of a major sugar chain of subunit β was presumed to be a xylomannose-type sugar chain, M3X. |
