Five-week-old rats were fed on a diet containing phosphorus either at 0.3% (CD) or at 1.5% (HD) for 21 days. Mitochondrial fraction (MF) was obtained from the kidney by the usual method. Contents of calcium, magnesium and phosphorus in MF obtained from rats fed HD were significantly higher than those of CD. We investigated whether elevated mitochondrial calcium of rats fed HD induces changes in the oxidative phosphorylation system. Respiration of kidney mitochondria of rats fed HD was not induced by ADP. Mitochondrial dehydrogenase activity of rats fed HD induced by 0.5mM succinate and 0.4mM ADP was lower than that of rats fed CD. Mitochondrial respiration with NAD^+ -linked substrates like pyruvate, glutamate or malate was not induced by 0.4mM ADP in rats fed HD. Activity of complex II, Mg-dependent ATP-ase and isocitrate dehydrogenase of mitochondria of rars fed HD were lower than those of rats fed CD. The low activities of NADH reduction, complex II and ATPase in rat fed HD may cause to the lack of mitochondrial respiration. |
