Three kinds of subunits, I, II and III, were separated from OAI-1 under a denaturing condition by size-exclusion HPLC and their N-terminal 20 amino acid sequences were analyzed. Subunits I and II of OAI-1 were shown to have the same sequences as subunits I and II of OAI-2 have, respectively. On the other hand, subunit III was a newly isolated subunit in this study and was not obtained from OAI-2. N-terminal 20 amino acid sequences of subunits II and III of OAI-1 were almost identical but the 10th cycles were different, that is glycine for subunit II and alanine for subunit III. The 12th cycles of the subunits II and III were not identified but seemed to be asparagine with a sugar chain. Differences in properties of OAI-1 and OAI-2 were believed to be due to the difference in quaternary structures of the proteins. |
